Protocols
Standard operating procedures for SCOPE detection reagents and reference materials. All protocols are optimized for reproducible flow cytometry results.

Reagents
- SCOPE CAR Detection Reagent (3 µL per test)
- Fluorochrome-conjugated backbone antibodies (e.g., CD45, CD3)
- FACS staining buffer (any commercially available)
- PBS (for final resuspension)
Materials
- Suitable staining tubes (e.g., 5 mL polystyrene FACS tubes)
- Centrifuge capable of 300 × g
- Micropipettes and tips
- Flow cytometer
Procedure
- 1Count cells and prepare 1 × 10⁶ PBMCs per test.
- 2Add 2 mL FACS staining buffer (any commercially available staining buffer), mix gently.
- 3Centrifuge cells at 300 × g for 5 minutes and discard supernatant.
- 4Resuspend the cell pellet in 100 µL FACS staining buffer.
- 5Add 3 µL SCOPE CAR Detection Reagent.
- 6Add fluorochrome-conjugated backbone antibodies as required (e.g., CD45, CD3).
- 7Mix gently by pipetting or brief vortexing.
- 8Incubate for 15 minutes at room temperature, protected from light.
- 9Add 2 mL FACS staining buffer, mix gently.
- 10Centrifuge at 300 × g for 5 minutes and discard supernatant.
- 11Repeat the wash once more with 2 mL FACS staining buffer and centrifuge again at 300 × g for 5 minutes.
- 12Discard supernatant and resuspend cells in 200 µL PBS.
- 13Proceed directly to flow cytometric acquisition and analysis.
Notes
- All steps should be performed at room temperature unless otherwise specified.
- Protect samples from light after adding fluorochrome-conjugated reagents.
- Adjust antibody volumes according to manufacturer recommendations and titration results.
